Nicotinamide riboside is a new generation of vitamin B3 (niacin or nicotinic acid). Nicotinamide riboside is a water-soluble vitamin (B3) that plays a role in over 200 biochemical reactions throughout the body. It is the beneficiary of several health claims. For example, it promotes energy production, contributes to the proper functioning of the nervous system and normal psychological functioning. Vitamin B3 is also important for the metabolism of fats, carbohydrates and proteins.

Precursor to coenzyme NAD+
The body uses vitamin B3 as a raw material for the coenzyme NAD+. The difference between nicotinamide riboside and other forms of vitamin B3 is that nicotinamide riboside is an exceptionally efficient precursor to NAD+ (nicotinamide adenine dinucleotide). As a natural substance, nicotinamide riboside is present in milk, though only in minimal trace amounts.
Powerful flush-free effect
There are different forms of vitamin B3: niacin (or nicotinic acid) and niacinamide (or nicotinamide riboside). Niacin may cause hot flushes after ingestion: the skin and face become warm and red. This may be accompanied by an annoying stinging, burning and itchy feeling. Nicotinamide riboside does not cause this flush by-effect despite its having very powerful intended effects.
Nicotinamide riboside is also called the 'miracle molecule' because it is the most powerful NAD+ booster. NAD+, or nicotinamide adenine dinucleotide, is an essential molecule found in every cell. As a coenzyme, NAD+ ensures that sugars, proteins and fats are converted into energy within the cell. This usable energy comes in the form of electrons. NAD+ is a biochemical electron carrier and is therefore the most important substance in the functioning of mitochondria, the cell’s power plants.
Substrate for NAD+ consuming enzymes
NAD+ is also an important substrate for a variety of NAD+ consuming enzymes, including PARP and sirtuin enzymes. Sirtuin enzymes are also activated by resveratrol, fasting and calorie restriction, among other factors. Although the molecule NAD+ has been known to scientists for over a century, it is increasingly being analyzed. It has been found that NAD+ may decrease with age and due to metabolic stress. Supplementation with the precursor to NAD+, nicotinamide riboside, may therefore be highly eligible in certain situations.

Nicotinamide riboside or nicotinamide mononucleotide?
Both nicotinamide riboside (NR) and nicotinamide mononucleotide (NMN) are precursors to NAD+. However, the NMN molecule is larger than the NR molecule. NMN must therefore first be broken down before it can make NAD+. NR has direct access to the cell and is a form of vitamin B3; NMN is not. Finally, NR is the beneficiary of 3 American and 1 European safety notifications and is made by the American company Chromadex, in full compliance with the standards of Good Manufacturing Practice (GMP).
